Analysis of air for microbiology is done by :
1) Setting place technique :
Expose plates containing sterile nutrient agar for 15 min in different location as follows.
1] Laminar flow bench.
2] Stainless steel table in exotic room.
3] Microbiology-lab
4] Pharmaceuticals Manufacturing Aria.
5] Incubate the plates at 370c for 48hrs. Determine the no. of colony forming units (C.F.U.)
2) Liquid impingement technique:-
Take 200 ml of sterile nutrient broth in a sterile conical flask. Using a suitable assembly such some our tho the sterile glass tubing into the medium, plate out 1,2 ml of the broth in sterile agar plates four nutrient agar mix well, allow to set and incubate at 370 for 48 hrs. Determine the no. of C.F.U. at the end of incubation.

How To determine the coli form count of water sample by most probable number (MPN) technique.
Principle: Coli form count of a water sample gives an idea of the of portability of water. The MPN technique is based on statistical sampling theory, which enables us to calculate the stable size replicative volume. Sample of 1000 count inoculated in to tubes of liquid medium gives a pattern of growth or no growth from which the count can be calculated.
Requirements: Lactose broth tubes containing Durham’s tube water sample, sterile pipettes, bromothymol blue indicator.
Procedure:- Inoculate water sample as follows :
a] 10ml(5ml) of water sample in each of stub, containing 10ml (5ml) of double strength lactose broth.
b] 1ml of water sample in each of 5 tubes containing 5ml of single strength lactose broth.
Incubate the tubes of 35-370c for 48hrs. and note the number of tubes showing acid gas product. Acidity can be checked with bromothymol blue indictor read coli form count from standard MPN tubes.
